本実習プログラムは、基礎的なスキルの習得と“考えるトレーニング”を目的とし、文理の垣根を越え少人数制にて実施しています。㋑クラシカルな分子生物学実験:PCR法を用いた身近な食品・生き物の種別判別実験、㋺自由課題:身近なモノを活用した時短・節約PCR法の開発等、基礎と展開を組み合わせて構成されています。実質6日間の集中講義形式で忙しい定期試験が終了した直後の夏休み(8月上旬)と春休み(2月上旬)に開講しています。

0. 本実習の履修を検討している学生さんたちへ

実験室で実際に手を動かして生命科学実験を体験することが本授業の特色の一つです。作業してみると、教科書やビデオ学習とは異なり、思わぬところで失敗(小さなつまずき)や気づきがあります。これらの良質な体験を通じた学習は、資料配布 + 質疑応答、オンデマンド授業 + 質疑応答、等の現状の代替手段では実施することは極めて困難であると考えています。そのため、本授業は対面授業に参加可能な学生のみに履修を制限します。また、きめ細かい指導質問・交流しやすい雰囲気づくり、も本授業のミッションとしています。そのため、履修希望者多数の場合は抽選します。

1. 実習の内容

第1部:分子生物学的手法の基礎体験 ・スキル習得

マグロの切り身もしくは、ブロックから、DNA抽出、PCR法による増幅、サンガーシーケンス法によるDNA配列決定、配列解析を行います。最終的に得られたDNA配列情報をデータベース参照により既知のマグロ配列と比較すします。その結果から、自分がどの種別のマグロ属の切り身を与えられたのかを考察し、ラボミーティングスタイルで結果考察を発表します。

③ 遺伝子増幅法(PCR法)によるDNAの増幅とDNA配列特異的切断酵素による切断
④ DNA断片を電気泳動により分離して検出
⑤ 得られたDNAのパターンからマグロの種別判別
⑥ サンガーシーケンス解析によるマグロの種別判別
⑦ 結果に関する発表および、ディスカッション 

第2部: “考えるトレーニング”

上記の既定の作業に加え、参加している学生各自の学習到達度・実験の進行度合いに個別に対応し、初心者でも理解可能なシンプルな課題を個人/グループに随時与えます。Web 検索や過去のプリント集を閲覧しても、答えは簡単には見つかりません。各自/グループは、実験を自らデザインして各種の検討を行う必要もあります。規定の実習の作業内容に追加するこの“考えるトレーニング” 、答えが確定していない課題に挑戦する体験楽しみながら実習に取り組んでください

2. ガイダンス

昼休み中、12:15~12:45 にオンライン(Zoom)にて実施します。Zoom URLは、UTASを確認してください。オンライン開催なので、昼食をとりながら参加しても構いません。実験室でガイダンスを受けたい、実験室を見学したい学生さんは、下記の問い合わせ先から担当教員へ連絡してください。

Experiments a lot & “thinking training”
Students in any division interested in life science are welcome.

It is complicated to classify tuna (sushi, fillets, blocks) by taste or shape unless you are a fish expert. So then, how do we determine the type of tuna accurately and reproducibly?

We will first receive a tuna fillet or sushi as a specimen in this practical course. Next, we will analyse the tuna using molecular biological methods to determine its tuna species. Finally, we will give a mini-presentation on our results in this course.

This course aims to learn (1) basic experimental skills and discussion methods and (2) an understanding of life science terms and techniques such as DNA and PCR, often mentioned in the daily news.

0. For students who are considering taking this course

We welcome participation by life science novices and non-science majors from any division.

Only students attending the face-to-face class are eligible to take the course.

If the number of students exceeds the capacity, there will be a selection process for those who can take the course. For details, please check 0-2.

1. Content of this course

Part 1 (A-side)

To Acquire skills that form the basis of molecular biological experiments.

Perform DNA extraction, amplification by PCR, DNA sequencing by Sanger sequencing method, and sequence analysis from tuna fillets or blocks. The final DNA sequence information obtained is compared with known tuna sequences by database reference. Based on the results, students will discuss which species of tuna genus they were given the fillets and present the results discussed in a lab meeting style.

  • Sequencing of PCR products by Sanger sequencing method

Part 2 (B-side)

“Thinking training”

In addition to the above flow, individuals/groups are given simple assignments that even beginners can understand, with individualised attention to the participating students’ learning achievement and experimental progress. We cannot easily find the answer by searching the Web or browsing past print collections. Also, each student/group needs to design and conduct various experiments.
“Thinking training” will be added to the work content of the prescribed practical flow, the experience of challenging tasks for which the answers are not fixed. We hope that you will enjoy your stay on this course.

Depending on the progress of the experiment, it may not be possible to complete the course at the following time.

Students who cannot attend the practical training day due to periodic examinations or other individual circumstances should be sure to notify us in advance. Please make individual schedule adjustments so as not to affect the progress of the practical course.
