格安顕微鏡でこれだけ見える、楽しい!
メダカが卵を産んだので、私が3年前に購入した格安顕微鏡で、どこまで観察できるか試してみました。身近なものを使って、セッティングを行いましたが、想像していたよりも、はるかに綺麗に撮影できました。魚類の発生生物学の専門家ではなく、生物学の素人でも手軽に楽しめる内容です。お金をかけずに身近な生命科学に触れたい人にも、参考になるかもしれません。
機材は全て私物(私費購入)です。また、この記事はDIYバイオロジーに興味を持つ人にも役立つ情報を提供することを目的としています。観察によって、みなさんにとってよい気づきがあると期待し公開します。

準備するもの

  • ワイヤレスデジタル顕微鏡, Amazon
  • Gakkenニューブロック, 学研
  • iPad, Apple
  • ワイヤーメッシュ, DAISO
  • フードコンテナ, MUJI
  • 偏光版, 東急ハンズ
  • モバイルバッテリー(長時間撮影時)
  • 自転車用LEDライト

手順

  1. Gakkenニューブロックとワイヤーメッシュを写真のように組み立てる。
  2. ワイヤレスデジタル顕微鏡を写真のように設置する。
  3. ワイヤレスデジタル顕微鏡のスイッチを入れる。
  4. iPadのアプリ, Max-seeを開く。
  5. アプリとワイヤレスデジタル顕微鏡が同期していることを確認する。
  6. ワイヤーメッシュの上に、メダカの卵が入っているフードコンテナをおく。そのとき、ワイヤレスデジタル顕微鏡のレンズの上に、卵が来るように、上から目視で確認する。
  7. ワイヤレスデジタル顕微鏡のステージを動かして、フォーカスがある場所を見つける。このときは、倍率約1000xに設定。
  8. iPad上でメダカの卵を観察できることを確認。静止画、動画を目的に応じて撮影。
  9. 観察セットの上部から、白色LED(自転車用)で卵をいろんな角度から照らすと、様々な見え方して面白い。
  10. 観察が終わったら、メダカの卵が入ったフードコンテナを安全な場所に戻し、また観察するまで保管する。

注意点・コメント

  • メダカの卵が入ったフードコンテナをこぼさないように、卵にとっても、iPadやモバイルバッテリーにとってもよくないです。
  • 撮影セットのブロックは、保育園で使っていたモノです。身の回りで代用できるものを自分で探すことも楽しいですよね。ブロックは一生物であることがわかりました。
  • タッパーは、普段おやつを入れている物で代用しました。メダカに使用後は、メダカ専用のタッパーにしています。
  • 偏光フィルターは、授業「茶わんの湯」で、鳥井寿夫先生に使いたかを教わり、光と撮影に関するテクニックを応用しています。光を正しく理解すると、素敵な写真と動画が撮れる。

Even with an inexpensive microscope, It’s fun.
A killifish laid eggs, so I tried to see how far I could observe them with a cheap microscope I bought three years ago. I set it up using what I had around me, and it turned out to be much clearer than I had imagined. It is easy and enjoyable for non-specialists in fish developmental biology and even for amateurs in biology. It will also be of interest to those who want to experience life science in an accessible way without spending a lot of money. All equipment is personal (purchased at my own expense). This article is also intended to provide useful information for those interested in DIY biology. We hope that you will find something of interest through our observations.

What to Prepare

  • Wireless digital microscope, Amazon
  • Gakken New Block, Gakken

  • LED light for bicycle

Procedures

  1. Assemble the Gakken New Block and wire mesh as shown in the photo.
    Set up the wireless digital microscope as shown in the photo.
  2. Switch on the wireless digital microscope and open the iPad app, Max-see.
    Make sure that the app and the wireless digital microscope are synchronized.
    Place a food container containing killifish eggs on the wire mesh.
  3. Visually check from above that the eggs are aligned with the lens of the wireless digital microscope.
  4. Move the stage of the wireless digital microscope to find the focus point. At this point, set the magnification to approximately 1000x.
  5. Confirm that the medaka eggs can be observed on the iPad.
  6. Take still and moving images for your purposes.
  7. It is interesting to illuminate the eggs from various angles with a white LED (for bicycles) from the top of the observation set to see the eggs in different ways.
  8. When you are done observing, return the food container with the killifish eggs to a safe place and store it until you are ready to observe again.

Comments

  • Don’t spill the food container with the killifish eggs in it, it’s not good for the eggs, nor for your iPad or mobile battery.
  • The blocks in the photo set are from the nursery. It is fun to find your own substitutes for things around you. The blocks have lasted a lifetime. For the
  • Tupperware, I substituted what I usually use to store sweets. After using it for killifish, I use it as a special Tupperware for killifish.
  • The use of the polarizing filter was taught to me by Dr. Torii in the “Lesson from the Tea Cup“, and I applied the techniques related to light and photography. When you understand light correctly, you can take great photos and videos without expensive equipment.
